Overview
Aprotinin is an antiplasmin agent i.e. hemostatic. Aprotinin is a serine protease inhibitor. Aprotinin is used in acute pancreatitis (inflammation of pancreas), in the management of sudden bleeding episodes in hemophilias and other bleeding disorders.
Indications
Aprotinin is primarily indicated in conditions like Abnormal blood loss prevention, Undergoing cardiopulmonary bypass, Fat embolism syndrome, Hyperfibrionolytic hemorrhage, Hyperplasminaemia, Liver transplantation, Muscle relaxation (short to intermediate duration), Pancreatic and endotoxic shock, Pancreatitis, Ruptured intracranial aneurysm, Traumatic and hemorrhagic shock.
Contraindications
Aprotinin is contraindicated in conditions like Hypersensitivity.
Side Effects
The symptomatic adverse reactions produced by Aprotinin are more or less tolerable and if they become severe, they can be treated symptomatically, these include Hypersensitivity reactions.
Warnings
Aprotinin hypersensitivity should be tested prior to administration. All patients should receive a test dose to assess possible allergic reaction. Those who have had previous therapy with aprotinin should be treated with special caution to avoid a possible anaphylactic reaction. In patients being given a repeat dose of aprotinin, an HÀ-histamine antagonist can be administered before the loading dose. Safe use of aprotinin in children has not been established.
High Risk Groups
Drug should not be given to Paediatrics, Pregnant Mothers, and Neonates.If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.
Adult Dosage
1.66 ug/kg.min (1.7 (1.66)) As recommended. IV — Maintenance, As Required
2.1 to 2.8 mg/kg (2.4 (2.45)) As recommended. IV — Load for Once
