Overview
Phenoxybenzamine (HCl) is irreversible a-adrenergic blocking agent, with a non-selective, covalent binding with both a1 post-synaptic and a2 presynaptic receptors. Chemically related to nitrogen mustard. it has a long duration of action. Phenoxybenzamine (HCl) is used to control hypertension and sweating in pheochromocytoma, to treat Raynaud`s phenomenon, to treat micturition disorders resulting from neurogenic bladder dysfunction.
Indications
Phenoxybenzamine (HCl) is primarily indicated in conditions like Adjunct in hypertensive episodes in pheochromocytoma, Hypertension and can also be given in adjunctive therapy as an alternative drug of choice in Raynaud's phenomenon, Urinary obstruction.
Contraindications
Phenoxybenzamine (HCl) is contraindicated in conditions like Orthostatic hypotension, Fluid depletion.
Side Effects
The severe or irreversible adverse effects of Phenoxybenzamine (HCl), which give rise to further complications include Postoperative hypotension. Phenoxybenzamine (HCl) produces potentially life-threatening effects which include Postural hypotension. which are responsible for the discontinuation of Phenoxybenzamine (HCl) therapy. The symptomatic adverse reactions produced by Phenoxybenzamine (HCl) are more or less tolerable and if they become severe, they can be treated symptomatically, these include Dizziness, Lethargy, Sedation, Nasal stiffness, Nausea and vomiting, Miosis, Ejaculatory failure.
Warnings
Phenoxybenzamine should be used with caution in patients with marked cerebral or coronary arteriosclerosis or renal (kidney) damage.
High Risk Groups
Drug should not be given to Pediatrics, Pregnant Mothers, Geriatrics, and Neonates. If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.
Adult Dosage
0.142 to 1 mg/kg (0.57 (0.571)) 12 hourly PO — -
1 mg/kg (1 (1)) 24 hourly IV Inf — For atleast 2 hrs., As Required
Child Dosage
0.5 to 1 mg/kg (0.75 (0.75)) 12 hourly Oral — For Paeds of All Ages
