Polymyxin B (Sulphate) is contraindicated in conditions like Hypersensitivity to the drug.

WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Tell your doctor or get medical help right away if you have any of the following signs or symptoms that may be related to a very bad side effect: All injection products: Signs of an allergic reaction, like rash ; hives; itching ; red, swollen, blistered, or peeling skin with or without fever ; wheezing ; tightness in the chest or throat; trouble breathing, swallowing, or talking; unusual hoarseness; or swelling of the mouth, face, lips, tongue, or throat. Flushing. Dizziness. Diarrhea is common with antibiotics . Rarely, a severe form called C diff–associated diarrhea ( CDAD ) may happen. Sometimes, this has led to a deadly bowel problem ( colitis ). CDAD may happen during or a few months after taking antibiotics. Call your doctor right away if you have stomach pain, cramps, or very loose, watery, or bloody stools. Check with your doctor before treating diarrhea. Injection (if given into the spine): Fever. Headache . Stiff neck.

Before initiating therapy with polymyxin B determine baseline kidney function. Serum concentration must be monitored regularly during treatment. It should be used with caution in patients with renal impairment, myasthenia gravis or other condition of neuromuscular disease, in patients known to have hypersensitivity reactions, in pulmonary disease, if sign of respiratory distress or nephrotoxicity occur, the drug should be discontinued. Take appropriate measures in case of secondary infection.

Drug should not be given to Pregnant Mothers, patients suffering from Kidney dysfunction, and Neonates. If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.

1 stripe (1 (1)) 12 hourly Topical — Apply Ointment on effacted area. 10000 units/g (10000 (10000)) 12 hourly Ophthalmic,Otic — Ointment 15000 to 25000 units/kg/body weight/day (20000 (20000)) 24 hourly IV — Infusions may be given every 12 hours, however, the total daily dose must not exceed 25,000 units/kg/day. 15000 to 30000 units/kg/day (22000 (22500)) 24 hourly IM — This should be reduced in the presence of renal impairment.The dosage may be divided and given at either 4 or 6 hour intervals. 5 to 10 ml (7.5 (7.5)) 6 hourly Ophthalmic — Frequency may be increased upto 6 times/day.
