Teniposide is contraindicated in conditions like Hypersensitivity to the drug.

WARNING/CAUTION: Even though it may be rare, some people may have very bad and sometimes deadly side effects when taking a drug. Tell your doctor or get medical help right away if you have any of the following signs or symptoms that may be related to a very bad side effect: Signs of an allergic reaction, like rash; hives; itching ; red, swollen, blistered, or peeling skin with or without fever ; wheezing ; tightness in the chest or throat; trouble breathing, swallowing, or talking; unusual hoarseness; or swelling of the mouth, face, lips, tongue, or throat. Signs of infection like fever, chills, very bad sore throat , ear or sinus pain, cough , more sputum or change in color of sputum, pain with passing urine, mouth sores, or wound that will not heal. Signs of bleeding like throwing up or coughing up blood; vomit that looks like coffee grounds; blood in the urine; black, red, or tarry stools; bleeding from the gums; abnormal vaginal bleeding; bruises without a cause or that get bigger; or bleeding you cannot stop. Signs of high or low blood pressure like very bad headache or dizziness, passing out, or change in eyesight. Signs of too much acid in the blood ( acidosis ) like confusion; fast breathing; fast heartbeat ; a heartbeat that does not feel normal; very bad stomach pain, upset stomach, or throwing up; feeling very sleepy; shortness of breath ; or feeling very tired or weak. A fast heartbeat. A heartbeat that does not feel normal. Flushing. Feeling confused. Headache. This medicine may cause tissue damage if the drug leaks from the vein. Tell your nurse if you have any redness, burning, pain, swelling, blisters, skin sores, or leaking of fluid where the drug is going into your body.

Teneposide is a potent drug and should be used only by physicians experienced in the administration of cancer chemotherapeutic drugs. Blood counts as well as renal and hepatic function tests should be carefully monitored prior to and during therapy. Physicians should be aware of the possible occurrence of a hypersensitivity reaction variably manifested by chills, fever, urticaria, tachycardia, bronchospasm, dyspnea, hypertension or hypotension and facial flushing. This reaction may occur with the first dose of teneposide and may be life threatening if not treated promptly with antihistamines, corticosteroids, epinephrine, intravenous fluids and other supportive measures as clinically indicated. In all instances where the use of Teneposide is considered for chemotherapy, the physician must evaluate the need and usefulness of the drug against the risk of adverse reactions. Most such adverse reactions are reversible if detected early. If severe reactions occur, the drug should be reduced in dosage or discontinued and appropriate corrective measures should be taken according to the clinical judgment of the physician. Reinstitution of Teneposide therapy should be carried out with caution, and with adequate consideration of the further need for the drug and alertness as to possible recurrence of toxicity.

Drug should not be given to Pregnant Mothers.If prescribing authority justifies the benefits of the drug against the possible damages he/she should reevaluate them and consult the reference material and previous studies.

50 to 165 mg/m2 (110 (107.5)) 24 hourly IV — Initially for 5 days then twice weekly as a 30-60 minutes infusion solution of upto 1mg/ml in NaCl 0.9% or D5W.
